Overview
Kisspeptin is a family of neuropeptides encoded by the KISS1 gene and produced primarily in hypothalamic neurons that project to GnRH neurons. Its critical role in reproductive biology was established following the 2003 New England Journal of Medicine publication by Seminara et al. documenting that loss-of-function mutations in the GPR54 kisspeptin receptor caused idiopathic hypogonadotropic hypogonadism. This discovery established Kisspeptin as the gatekeeper of the GnRH pulse generator and the entire gonadotropin axis. How It Works
Kisspeptin activates GPR54 receptors on GnRH neurons in the hypothalamus, triggering pulsatile GnRH release into the portal circulation. This GnRH pulse then drives LH and FSH secretion from the anterior pituitary, stimulating gonadal steroid production and gametogenesis. Kisspeptin neurons integrate signals from sex steroids, metabolic status, and circadian inputs to modulate GnRH pulse frequency and amplitude.
